This work has been designed to develop a hydrogen fermentation process for Deoiled Jatropha Waste (DJW) and it has been divided in to 2 major parts, whereas the former involve the raw DJW and the later involve the pretreated DJW (hydrolysate). In order to check biohydrogen production potential of r

aw DJW, 5 seed sources were chosen, among them sewage sludge (from Chung Hsing municipal wastewater treatment plant) showed as a good harbor of hydrogen producing bacteria. Optimization of substrate concentration, temperature and pH was carried out using single factor at a time design and resulted i

n an optimized level of 200 g/L, 550C and 6.5. At these optimal conditions the hydrogen production rate (HPR) and hydrogen yield (HY) values were noted as 0.74 L/L-d and 10.6 mL/g Volatile Solids (VS) added. Monod model was used to elucidate the effect of substrate concentration. Various pretreatmen

t methods were adopted in order to enrich the hydrogen producing microbial population. Of all the methods employed heat treatment enhanced the production performance to 4 folds increment. Heat treatment at 950C for 30 minutes provided the HPR of 1.4 L/L-d and statistically determined values (using c

entral composite design- CCD design) also satisfactorily matched. With the optimal conditions attained an Anaerobic Sequencing Batch Reactor (ASBR) was operated and the effect of various hydraulic retention time (HRT) was studied. The results showed that 1.75 days HRT was efficient and provide a pea

k HPR of 1.45 L/L-d and HY of 9.7 mL/g VS added. The microbial insights accessed via PCR-DGGE (polymerase chain reaction- denaturing gradient gel electrophoresis) revealed that combination of cellulolytic and fermentative bacteria present in the reactor.Hydrogen production potential could be enhance

d via increasing the sugars which can be achieved via pretreatment methods. Various pretreatment and hydrolysis methods were employed in order to achieve the maximal sugar release from DJW. At initial screening, acid (HCl, 2%) pretreatment was proved as efficient. The acid unhydrolysed biomass (AUHB

) was pretreated by enzyme (Viscozyme, 5%). Combining acid and enzyme pretreatment have achieved the maximal sugar recovery of more than 90% based on the cellulose (cellulose+ hemicellulose) content. Batch hydrogen fermentation of the hydrolysates obtained was carried out. The results proved that ac

id and enzyme pretreated hydrolysates were suitable for the hydrogen generation. Increasing biomass concentration has increased the amount of releasing sugars. The biomass concentration of 100 g/L (for acid hydrolysis) and 200 g/L (for enzyme hydrolysis) were fixed as optimal for the hydrolysate pre

paration. The structural modifications occurred during the pretreatment were explained via FTIR, XRD and BET analysis. Optimization studies showed that optimal temperature and initial pH as 370C and 5.5 for pretreated hydrolysate. Two Continuously stirred tank reactors (CSTR) were started with the h

ydrolysates pretreated via acid (HCl, 2%) and enzyme (Viscozyme, 5%). Cell washout occurred due to the cells in suspension form at low HRT of 24 hrs. However, a novel hybrid immobilized cell system has been developed and implemented to overcome such problem. The batch results revealed that the immob

ilized cells could be used for more than 10 cycles proving the stability. The results of the immobilized cell reactor were turned out with a peak HPR and HY of 0.8 L/L*d and 81 mL/g Reducing Sugars (RS) added for acid hydrolysate and 3.7 L/L*d and 138 mL/g RS added for enzyme mediated hydrolysate wh

ile the optimal HRTs were 16 and 12 hrs, respectively. The microbial community analysis revealed that majority of the PCR-DGGE bands represent the hydrogen producers (Clostridia spp.) reported previously. FISH results also supported that the bacterial population mostly comprised of Eu bacteria.A com

prehensive report provided the information about advantages and disadvantages of the raw and pretreated DJW fermentation process. Besides, the energy calculations revealed the efficiency of developed process. Additionally, CO2 emission reduction was also calculated. Gas separation using polyimide me

mbrane (UBE module) was performed for the purification and enrichment of H2. The results showed that optimization of gas composition, temperature and retentate/ feed flow ratio are important factors in the separation process. The selectivity (1.22-1.63) of the tested material is comparable with the

other commercial membranes. Integrating such gas separation system with the DJW fermentation system could make the process viable and sustainable towards the commercialization.
