醣分子位置選擇性保護的方法學開發和醣予體-受體反應性於醣鏈結反應的評估及統計

摘要化學醣鏈結反應是獲取高純度及高產量的目標多醣體並用於進行生物學評估的最有效方法之一。然而,在醣予體與醣受體鍵結時的區域選擇性及控制新醣苷鍵的立體選擇性依然是醣基化反應恆久的難題。在本論文的第一章總結了目前用於解決相關挑戰的各種重大發現。而第二章中,我們發現醣在使用三氟甲磺酸三甲基矽酯(TMSOTf) 催化下的非變旋異構羥基乙醯化選擇性,並能能保留住變旋異構中心的三甲基矽酯(O-TMS)保護基。與傳統的方法不同,1-O-TMS的醣基單元可以進而直接進行後續的醣基化反應,不需要在對變旋異構羥基進行修飾。因此能省略了攏長的保護-去保護反應,減少產物與時間的損耗,進而更簡便的進行化學醣鏈結反應。

在第三章中,我們探討了數組帶有α1,4 及β1,4不同內醣苷鍵鍵結的雙醣硫苷醣予體異構物,並檢測其反應性,發現前者的反應性明顯大於後者。這項差異可以根據立體電子效應解釋,也就是雙醣中的變旋異構及外變旋異構效應;此外,在統計各類雙醣與不同親核性的醣受體反應後的立體選擇性分析,發現到選擇性大致取決於本身雙醣中的醣苷鍵類型,而非非還原端醣上的取代基。因此相較於有著β1,4鍵結的雙醣,有著α1,4鍵結的雙醣在醣鏈結反應後表現出更佳的α-選擇性。同時,我們也發現這些雙醣在醣基化反應的產率更受到的醣受體親核性影響,雙醣醣予體的反應性影響則較小。關鍵字選擇性乙醯化, per-O-TMS 醣, 內醣苷鍵, 立

  從過去到現在,國內的休閒旅遊,已經漸漸吹起休閒林場的趨勢,透過接觸大自然、自然生態遊憩,紓解壓力、放鬆身心,已經變成生活在城市裡的人所憧憬的新樣貌生活。到目前為止,也有很多農家轉型成功變為觀光休閒林場,綜合遊樂、教育、觀光、休閒成為豐富多元化的新遊憩地點。所以,怎麼使傳統農場將規則化的農業生產、樸實的農村生活、好玩的遊憩觀光以及保育生態的功能,並且能與休閒遊憩活動緊密連結,以提升林場的收入經營,也使民眾的休閒旅遊質量升高,真的是作為農場未來發展的重要關鍵。  此研究旨在探究遊客的旅遊動機、知覺價值、幸福感與重遊意願關係的互相影響情形及差異,採用發放問卷進行資料蒐集並參閱相關文獻,分析工具

是根據SPSS18.0統計軟體,以t檢定、信效度分析、描述性統計分析、迴歸分析、單因子變異數分析、等進行數據統計分析。  研究結果顯示:旅遊動機對幸福感有部分顯著影響;知覺價值對幸福感有部分影響;旅遊動機對重遊意願有部分顯著影響;知覺價值對重遊意願有顯著影響;幸福感對重遊意願有顯著影響;旅遊動機、知覺價值、幸福感及重遊意願各構面間有顯著影響。根據上述研究結果,提供東勢林場經營管理者在設計林場旅遊時,能吸引更多的旅遊觀光潮流,且提升旅客的重遊意願。也希望依據研究結果提供建議,給予東勢林場作為之後經營管理改善的根據。
